INTRODUCTION
Aflatoxin is a Group 1 carcinogen and is a product of mould. Ochratoxin is a mycotoxin produced by the fungus Aspergillus ochraceous and also by several species of Penicillium fungi. It is considered a potential carcinogen and has been known to cause kidney damage. All these mycotoxins are monitored worldwide and allowed concentration limits depend not only on the territory, but also on the food/feed matrix and are given by local administrations.
ABSTRACT
HPLC/FLD determination of aflatoxin B1 (CAS 1162-65-8), B2 (CAS 7220-81-7), G1 (CAS 1165-39-5), G2 (CAS 7241-98-7) and ochratoxin A (CAS 303-47-9) on an ARION® Polar C18, 3.0 µm column (150 mm × 4.6 mm, ARI-5721-IK46). Gradient elution with A: 0.5 % acetic acid / 5 % ACN in water, B: methanol and C: ACN at 1.0 ml/min and 40 °C, injection 20 µl, fluorescence detection after post-column derivatisation (excitation 364 nm switched to 330 nm at 15 min, emission 455 nm, PMT gain 14). The four aflatoxins elute between 8 and 12 minutes and ochratoxin A at about 20 minutes. The pepper sample was cleaned up by means of a Vicam immunoaffinity column.
