The accurate, simple, and selective HPLC/UV method has been established and validated for the determination of the antibiotic AMP as a suitable method in a cardiac surgery which does not have to be used only in cardiac surgery. The new ARION® Polar C18 column has been shown to effectively separate AMP in human blood plasma.
ABSTRACT
HPLC/UV method for the determination of ampicillin (AMP, CAS 69-53-4) in human blood plasma, developed for dosage monitoring in cardiac surgery, on an ARION® Polar C18, 5.0 µm column (250 mm × 4.6 mm, ARI-5721-LM46). Isocratic elution with 15 mM KH2PO4 buffer pH 3.3 / MeOH 75/25 (v/v) at 1.4 ml/min and 30 °C, injection volume 25 µl, UV detection at 210 nm, with cephalexin as internal standard. Ampicillin elutes at about 10.5 minutes. Developed by the Department of Pharmacology, Faculty of Medicine and Dentistry, Palacky University Olomouc.
